En Cambridge colaboró con Crick en la investigación de la estructura del ADN y en 1953 adelantó que los componentes esenciales de este ácido -cuatro bases orgánicas- debían estar enlazados por pares: adenina con timina y guanina con citosina. Así pudo postular con Crick el modelo molecular de doble hélice para el ADN, en que una molécula podía duplicarse, puesto que las dos cadenas de la hélice eran complementarias.
James Dewey Watson trabajó luego en el Instituto Tecnológico de California, en Pasadena (1953-55) y en la Universidad de Harvard, donde llegó a ser profesor de bioquímica y biología molecular (1961). Finalmente ayudó a descifrar el «código genético» contenido en las secuencias del ADN y descubrió el ARNm (ácido ribonucleico mensajero), que transfiere el código ADN a las estructuras celulares formadoras de proteínas.
